Mass to Moles Conversion Formulas

1. Calculate Moles of RNA

moles of RNA [mol] = RNA mass [g] / RNA molecular weight [g/mol]

This equation converts the physical mass of RNA to the number of moles using the molecular weight.

2. Calculate Moles of RNA Ends

moles of ssRNA ends [mol] = moles of ssRNA [mol] × 1

Each single-stranded RNA molecule has one 3' and one 5' end counted as one end unit for ligation calculations.

3. Calculate RNA Copy Number

RNA copy number [molecules] = moles of RNA [mol] × 6.022e23 [molecules/mol]

Uses Avogadro's number to convert moles to actual number of RNA molecules.

4. Calculate RNA Molecular Weight

RNA molecular weight [g/mol] = (RNA length [nt] × average nucleotide weight [g/mol per nt]) + terminal correction [g/mol]

When sequence is provided, the exact nucleotide weights are summed. When only length is provided, an average nucleotide molecular weight (~325 g/mol) is used.
